Proceptin 40Â
Presentation: Proceptin 40 is available as enteric-coated capsules containing 40 mg of omeprazole.Â
Indications: Proceptin 40 (omeprazole) is indicated for the treatment of conditions caused by excessive stomach acid production, including:
- Duodenal ulcers
- Gastric ulcers
- NSAID-associated duodenal and gastric ulcers
- Prophylaxis in patients with a history of NSAID-associated duodenal and gastric ulcers
- Gastro-esophageal reflux disease (GERD)
- Long-term management of acid reflux disease Â
- Acid-related dyspepsia Â
- Severe ulcerating reflux esophagitis Â
- Prophylaxis of acid aspiration during general anesthesia Â
- Zollinger-Ellison syndrome Â
- Eradication of Helicobacter pylori in combination with antibiotics.
Dosage and Administration: The dosage of Proceptin 40 will vary depending on the condition being treated and the patient's response, as determined by a physician.
- Oral Administration:
- Benign gastric and duodenal ulcer: 20 mg once daily for 4 weeks (duodenal ulcer) or 8 weeks (gastric ulcer). In severe or recurrent cases, the dose may be increased to 40 mg daily. Maintenance dose for recurrent duodenal ulcer is 20 mg once daily; for prevention of relapse, 10-20 mg daily.
- NSAID-associated duodenal or gastric ulcer: 20 mg once daily for 4 weeks, continued for a further 4 weeks if not fully healed. 20 mg once daily is used as prophylaxis in patients with a history of NSAID-associatedulcers.
- Gastro-esophageal reflux disease: 20 mg once daily for 4 weeks, continued for a further 4-8 weeks if not fully healed; 40 mg once daily may be given for 8 weeks in refractory cases; maintenance dose is 20 mg once daily. Â
- Long-term management of acid reflux disease:10-20 mg daily. Â
- Acid-related dyspepsia:10-20 mg once daily for 2-4 weeks. Â
- Prophylaxis of acid aspiration:40 mg on the preceding evening, then 40 mg 2-6 hours before surgery. Â
- Zollinger-Ellison syndrome:Initially 60 mg once daily; usual range 20-120 mg daily (ifthe daily dose is more than 80 mg, it should be divided and given twice daily).
- Helicobacter pylori eradication: 20 mg twice daily in association with specific antimicrobial agents (e.g., amoxicillin and metronidazole, or clarithromycin and metronidazole, or amoxicillin and clarithromycin) for one week.
- Intravenous Injection:
- Prophylaxis of acid aspiration:40 mg to be given slowly (over a period of 5 minutes) as an intravenous injection, one hour before surgery. Â
- Duodenal ulcer, gastric ulcer or reflux oesophagitis:In patientswhere oral medication is inappropriate, 40 mg once daily is recommended.
- Zollinger-Ellison syndrome: The recommended initial intravenous dose is 60 mg daily. Higher daily doses may be required and adjusted individually. When doses exceed 60 mg daily, the dose should be divided and given twice daily.
- Proceptin capsules should be swallowed whole and taken on an empty stomach, at least 1 hour before a meal.
Side Effects: Omeprazole, the active ingredient in Proceptin 40, is generally well-tolerated. Common side effects may include headache, diarrhea, nausea, vomiting, flatulence, abdominal pain, and constipation.
Less common side effects may include dizziness, dry mouth, insomnia, skin rash, and changes in liver enzymes. Long-term use may be associated with an increased risk of bone fracture, vitamin B12 deficiency, and hypomagnesemia.
Precautions: Before taking Proceptin 40, inform your doctor if you have any liver problems. Gastric malignancy should be excluded before starting treatment. Caution should be exercised during pregnancy and breastfeeding. Proceptin 40 may interact with other medications, including certain antifungals (e.g., ketoconazole, itraconazole), anticoagulants (e.g., warfarin), antiplatelet drugs (e.g., clopidogrel), and some HIV medications. Long-term use may require monitoring.
Storage: Store Proceptin 40 capsules below 30°C in a dry place, protected from light and moisture. Keep out of reach of children. For the intravenous injection, store as directed on the vial.
