Denixil Rapid 0.5 Tablet

Description

Denixil Rapid 0.5 Tablet contains clonazepam, a benzodiazepine, and is used to treat several conditions. Clonazepam has anticonvulsant, sedative, muscle-relaxing, and anxiolytic properties. It is effective in managing panic disorder, with or without agoraphobia, characterized by unexpected panic attacks and associated concerns. It is also indicated for Lennox-Gastaut Syndrome (petit mal variant), akinetic, and myoclonic seizures. Additionally, it may benefit patients with absence seizures (petit mal) who have not responded to succinimides.

Pharmacology

Clonazepam enhances the activity of GABA, a neurotransmitter that reduces nerve transmission in the brain. This mechanism is thought to be responsible for its ability to suppress spike and wave discharges associated with absence seizures. Its efficacy in panic disorder is not fully understood, but it is believed to involve interactions with serotonin and benzodiazepine receptors.

Dosage & Administration

The initial dose for adults with panic disorder is 0.25 mg given in two divided doses. An increase to the target maintenance dose of 1 mg/day may be made, as the patient tolerates. Increases should be in increments of 0.125 mg to 0.25 mg/day, every 3 days until the panic disorder is controlled or the maximum recommended daily dose of 4 mg is reached.

The elderly are particularly sensitive to benzodiazepines, and the usual adult initial dose should be used with caution, and dosage increments should be made more gradually.

Contraindications

Clonazepam is contraindicated in patients with:

  • A history of hypersensitivity to benzodiazepines.

  • Clinical or biochemical evidence of significant liver disease.

  • Acute narrow-angle glaucoma.

Precautions & Warnings

Withdrawal symptoms may occur following abrupt discontinuation of clonazepam. Long-term use of clonazepam may lead to drug dependence. Caution is advised when prescribing this medication to patients with a history of seizures and also in patients where a drop in blood pressure might lead to cardiac or cerebral complications.

Side Effects

The most frequent side effects are those related to CNS depression. Experience has shown that patients taking clonazepam commonly become somewhat drowsy.

Other side effects, seen less frequently, are:

  • Ataxia

  • Behavioral problems

  • Confusion

  • Constipation

  • Depression

  • Diarrhea

  • Dysarthria

  • Diplopia

  • Fecal incontinence

  • Headache

  • Increased salivation

  • Lassitude

  • Lightheadedness

  • Memory disturbance

  • Muscular weakness

  • Nausea

  • Nocturia

  • Pain

  • Palpitations

  • Paresthesia

  • Pneumonia

  • Respiratory depression

  • Rhinitis

  • Skin rash

  • Slurred speech

  • Urinary incontinence

Use in Pregnancy & Lactation

Clonazepam should only be used during pregnancy if the potential benefit outweighs the risk to the fetus. It should be used with caution in nursing mothers.
