Overview & Mechanism

  • Active Ingredient: Itraconazole (Triazole Antifungal)

  • Technology: Uses SUBA (Super Bio-available) technology to improve drug absorption and dissolution compared to standard crystalline forms.

  • Mechanism: Inhibits Cytochrome P-450 enzymes to block ergosterol synthesis, impairing fungal cell membrane growth and integrity.

Key Indications

  • Non-Systemic Infections: Oropharyngeal & vulvovaginal candidiasis, pityriasis versicolor, tinea infections (pedis, cruris, corporis, manuum), and onychomycosis.

  • Systemic Infections: Systemic candidiasis, aspergillosis, cryptococcosis (including cryptococcal meningitis), and histoplasmosis.

  • Prophylaxis: Maintenance therapy in AIDS patients and fungal prevention during prolonged neutropenia.

Dosage Highlights (Take with food)

  • Standard Dosing (100 mg / 200 mg):

    • Vulvovaginal Candidiasis: 200 mg twice daily for 1 day.

    • Pityriasis Versicolor: 200 mg daily for 7 days.

    • Tinea Infections: 100 mg daily for 15–30 days (or 200 mg daily for 7 days).

    • Systemic Diseases: 100 mg–200 mg once or twice daily depending on severity (duration ranges from weeks to months).

  • SUBA Dosing (65 mg / 130 mg): Requires reduced milligram dosages due to enhanced bioavailability (e.g., 65 mg daily/twice daily replacing standard 100 mg regimens).

Contraindications & Safety

  • Contraindicated In: Pregnancy, lactation, severe liver disease, and known hypersensitivity.

  • Major Interactions: Terfenadine, astemizole, cisapride, simvastatin, oral midazolam/triazolam, rifampin, phenytoin, digoxin, and calcium channel blockers.

  • Precautions: Antacids impair absorption—take indigestion remedies at least 2 hours after SUBA-i. Long-term use (>1 month) requires liver function monitoring.

  • Common Side Effects: Nausea, abdominal pain, headache, dizziness, elevated liver enzymes, and rash.
