Paloron 0.5 Tablet

Presentation: Each tablet contains 0.5 mg of Palonosetron as Palonosetron Hydrochloride USP. 

Indications: Paloron is an antiemetic medication used in adults and children (1 month to 17 years) to prevent and treat:

  • Nausea and vomiting caused by chemotherapy (both acute, occurring on the day of treatment, and delayed, occurring days after).
  • Nausea and vomiting caused by radiation therapy.
  • Nausea and vomiting after surgery.

Dosage and Administration: The usual adult tablet dosage is 0.5 mg daily.

For chemotherapy-induced nausea and vomiting:

  • Adults: 0.5 mg tablet approximately 1 hour before the start of chemotherapy.
  • Children (1 month to 17 years): A single intravenous (IV) dose at 20 mcg/kg body weight, with a maximum dose of 1.5 mg, given approximately 30 minutes before the start of chemotherapy.

For radiotherapy-induced nausea and vomiting:

  • Adults: A single IV dose of 0.25 mg should be administered over 30 seconds approximately 30 minutes before each week of radiation fraction.

For post-operative nausea and vomiting:

  • Adults: A single IV dose of 0.075 mg should be administered over 10 seconds immediately before the induction of anesthesia. 

Paloron tablets are for oral use. Follow your doctor's instructions carefully.

Side Effects: The most common side effects reported with Paloron tablets are:

  • Headache
  • Constipation

Less common side effects may include abdominal pain, fatigue, dizziness, and insomnia.

Precautions:

  • Inform your doctor if you have any known allergies to palonosetron or any of its ingredients.
  • Use with caution in patients who have exhibited hypersensitivity to other 5-HT3 receptor antagonists.
  • It is not known whether Palonosetron is excreted in breast milk, so caution should be exercised when administered to a nursing woman.
  • No dosage adjustment is generally recommended for elderly patients or those with kidney or liver problems.

Contraindications: Paloron is contraindicated in patients known to have hypersensitivity to palonosetron or any of its components.

Drug Interactions: In vitro studies suggest that Palonosetron is unlikely to have significant drug interactions with other medications metabolized by major CYP enzymes. Palonosetron has been safely administered with corticosteroids, analgesics, antiemetics, antispasmodics, and anticholinergic agents in clinical trials. 

Storage: Store below 30°C in a dry place, protected from light. Keep out of reach of children.
